Pearl-102 is a small molecule combination therapy being developed by PearlsInMires for the treatment of gastrointestinal (GI) solid tumors. The drug candidate is designed to target Poly(ADP-ribose) glycohydrolase (PARG) and Schlafen 11 (SLFN11). PARG is a critical enzyme in the DNA damage response, responsible for degrading poly(ADP-ribose) (PAR) chains; its inhibition leads to PAR accumulation, replication stress, and cell death. SLFN11 is a DNA/RNA helicase that sensitizes cells to DNA-damaging agents by blocking replication forks. Pearl-102 is currently in the IND-enabling phase, with the company aiming to complete these studies by 2026.
