Pearl-111 is a small molecule combination therapy being developed by PearlsInMires for the treatment of gastrointestinal (GI) solid tumors. The drug is designed to target Schlafen 11 (SLFN11) and the ADP-ribose pathway, which are critical components of the cellular DNA damage response. SLFN11 expression is a recognized biomarker that sensitizes cancer cells to DNA-damaging agents, and by co-targeting these pathways, Pearl-111 aims to enhance therapeutic efficacy and overcome resistance in solid tumors. As of 2024, the program is in the IND-enabling stage, with the company projecting the completion of IND-enabling tests by 2026.
