Drug intelligence / Profile preview
**Peatonide** is an oral nutraceutical fixed-dose combination of hydrodispersible palmitoylethanolamide and melatonin, containing 1,200 mg palmitoylethanolamide and 0.2 mg melatonin per daily bedtime dose. It has been investigated as preventive therapy for low-frequency episodic migraine. Palmitoylethanolamide is proposed to reduce neuroinflammatory signaling, including through peroxisome proliferator-activated receptor alpha activation and reduced mast-cell degranulation, while melatonin may support circadian regulation and modulate migraine-associated neuroinflammation. In a randomized, double-blind, placebo-controlled Italian study of 60 adults treated for three months, Peatonide was associated with reductions in monthly migraine days, attack duration, pain intensity, disability, and associated symptoms; larger multicenter studies are needed to confirm these findings.
