Overview

PEEL-224 is a novel small molecule nanoparticle topoisomerase I inhibitor developed for the treatment of advanced and refractory solid tumors, including pediatric cancers. The active molecule in PEEL-224 is derived from camptothecin, a natural compound found in the leaves and stem of *Camptotheca acuminata* (the "Happy Tree"), which has been used in traditional Chinese medicine. Through chemical optimization and nanocarrier assembly, PEEL-224 aims to improve tumor delivery and reduce toxicity compared to conventional chemotherapy agents like irinotecan. Preclinical studies have shown that it can overcome chemoresistance, with promising activity even against drug-resistant tumors. It is currently being evaluated as monotherapy and in combination with vincristine and temozolomide for various solid tumors—including neuroblastoma, Ewing sarcoma, rhabdomyosarcoma, desmoplastic small round cell tumor (DSRCT), osteosarcoma, primitive neuroectodermal tumors (PNET), peripheral neuroectodermal tumors (PNET), small cell sarcoma—and other aggressive pediatric cancers[1][2][3][5][6][7][8].
