PEEL-224 + FOLF + B is an investigational combination therapy consisting of PEEL-224, 5-fluorouracil (5-FU), leucovorin (folinic acid), and bevacizumab. Developed by Peel Therapeutics, this regimen is primarily being evaluated for the treatment of advanced solid tumors, with a focus on colorectal cancer and sarcomas. PEEL-224 is a pegylated topoisomerase I (TOP1) inhibitor, a polymer-drug conjugate derived from camptothecin designed to enhance the delivery and efficacy of the TOP1 inhibitor while minimizing systemic toxicities like neutropenia and diarrhea. In this combination, PEEL-224 acts as the cytotoxic TOP1 inhibitor, 5-FU and leucovorin provide thymidylate synthase inhibition, and bevacizumab targets vascular endothelial growth factor (VEGF) to inhibit angiogenesis. The combination is currently undergoing Phase 1 clinical trials to determine safety, tolerability, and preliminary efficacy.
