peg-8-psilocin is a non-psychedelic, PEGylated derivative of psilocin currently under preclinical investigation for the treatment of metabolic dysfunction-associated steatotic liver disease (MASLD). By conjugating psilocin with polyethylene glycol (PEG-8), the compound is engineered to limit central nervous system (CNS) exposure, thereby preserving peripheral serotonergic activity while minimizing psychoactive effects. In preclinical models of diet-induced MASLD, peg-8-psilocin has demonstrated the ability to reduce liver steatosis, limit body weight gain, and improve glucose and lipid homeostasis. It acts as a serotonin receptor agonist, specifically targeting peripheral 5-HT receptors to induce beneficial remodeling of the hepatic lipidome, improve insulin sensitivity, and restore pancreatic beta-cell ultrastructure.
