Overview

PEG-ADA2-E182T is a pegylated, engineered recombinant human adenosine deaminase 2 (ADA2) variant developed by Halozyme Therapeutics. It was specifically designed with an E182T mutation to eliminate enzymatic activity, serving as a negative control in preclinical studies investigating the role of adenosine depletion in the tumor microenvironment. While active PEG-ADA2 variants (such as PEG-ADA2-K374D) demonstrated significant tumor growth inhibition by reducing immunosuppressive adenosine levels, PEG-ADA2-E182T showed no anti-tumor activity, confirming that the therapeutic effect of this class of immunotherapies is dependent on ADA2 enzymatic function.
