Drug intelligence / Profile preview
PEG-AVP04 (also known as PEG-AVP0458 or 124I-PEG-AVP04 when radiolabeled) is a PEGylated, bivalent diabody (Avibody) developed by Avipep that targets tumor-associated glycoprotein 72 (TAG-72). Engineered to optimize pharmacokinetics and tumor bioavailability, the agent consists of two single-chain variable fragments (scFv) with surface disulfides precisely conjugated to PEG molecules to prevent rapid renal clearance while maintaining high target affinity. When radiolabeled with Iodine-124 (124I), PEG-AVP04 serves as a diagnostic PET imaging agent. It has been evaluated in Phase 1 clinical trials for the detection and biodistribution mapping of TAG-72-positive relapsed or metastatic prostate and ovarian cancers, demonstrating rapid tumor penetration, prolonged retention, and minimal uptake in normal tissues.
