Drug intelligence / Profile preview
PEG-CZNPs (phospholipid-polyethyleneglycol-capped ceria-zirconia nanoparticles) are antioxidant nanoparticles designed to restore impaired autophagy flux and scavenge reactive oxygen species (ROS). Developed by researchers at Konyang University and Hannam University, these ultrasmall (2-3 nm) nanoparticles utilize the redox cycling of cerium (between Ce3+ and Ce4+ states) stabilized by zirconia to reduce oxidative stress. PEG-CZNPs promote autophagy flux by inhibiting GSK3β, which enhances the nuclear translocation and activation of transcription factor EB (TFEB), and by restoring the blunted Akt/mTOR pathway. Preclinical studies in cellular and animal models have demonstrated their potential in treating Fabry disease by reducing intracellular globotriaosylceramide (Gb3) accumulation and alleviating kidney injury, as well as in mitigating obesity-induced organ damage and renal fibrosis.
