PEG-DAPD is a PEGylated dual-acting peptide developed for the treatment of type 2 diabetes. It functions by combining glucagon-like peptide-1 (GLP-1) receptor agonist activity with glucagon receptor antagonist activity. This dual mechanism aims to promote glucose-dependent insulin secretion from the pancreas and reduce glucose overproduction by the liver. The PEGylation of the original dual-acting peptide for diabetes (DAPD) was performed to overcome its short plasma half-life, resulting in dramatically prolonged in vivo activity. Additionally, PEGylation eliminates the inhibition of gastrointestinal motility observed with GLP-1 and its analogues, offering a potential therapeutic advantage for type 2 diabetes.
