Drug intelligence / Profile preview
A **combination therapy** of **peg-irinotecan** (also known as **EZN-2208**, which is pegylated SN-38, the active metabolite of irinotecan) and **cetuximab**. Peg-irinotecan is a small molecule chemotherapy agent designed for enhanced delivery and extended half-life via PEGylation, increasing exposure to the active metabolite SN-38 and improving efficacy in irinotecan-refractory tumors. Cetuximab is a chimeric monoclonal antibody that binds to and inhibits the epidermal growth factor receptor (EGFR), blocking downstream proliferation signals in tumor cells. The main investigated use is in patients with advanced or metastatic colorectal cancer (mCRC), especially after failure of standard therapies. The combination shows a similar safety and efficacy profile to irinotecan plus cetuximab, with potential benefit in irinotecan-resistant or -refractory cases[3].
