PEG-liposomal dexamethasone sodium phosphate (ENC-0313, Oncocort) is a pegylated liposomal formulation of the glucocorticoid dexamethasone sodium phosphate. Developed by Enceladus Pharmaceuticals, it consists of ~100 nm vesicles composed of dipalmitoyl phosphatidyl choline (DPPC), PEGylated distearoyl phosphatidyl ethanolamine (PEG-DSPE), and cholesterol. This formulation is designed to provide prolonged systemic circulation and enhanced accumulation at sites of inflammation or malignancy (via the enhanced permeability and retention effect) compared to conventional dexamethasone. It acts as a glucocorticoid receptor agonist. The drug has been evaluated in Phase I/IIa clinical trials for the treatment of metastatic castration-resistant prostate cancer (mCRPC) and multiple myeloma.
