**peginterferon alfa-2a + nucleos(t)ide analogues** is a combination regimen used primarily for the treatment of chronic hepatitis B virus (HBV) infection. Peginterferon alfa-2a is a pegylated form of interferon alpha-2a, an immunomodulatory biologic that enhances the host immune response against HBV, while nucleos(t)ide analogues (such as entecavir, tenofovir, lamivudine, adefovir, or telbivudine) are small molecules that inhibit HBV DNA polymerase, effectively suppressing viral replication. The combination exploits complementary mechanisms: the immunostimulatory action of peginterferon and the potent antiviral effect of NAs, theoretically aiming to increase rates of hepatitis B surface antigen (HBsAg) loss and functional cure. While combination therapy has shown increased on-treatment suppression of HBV and decline in HBsAg, evidence does not consistently demonstrate significantly higher rates of long-term HBsAg seroconversion or functional cure over monotherapy.
