peginterferon alfa-2a + ribavirin + amantadine

Peginterferon alfa-2a + ribavirin + amantadine is a combination therapy used primarily in the treatment of chronic hepatitis C, particularly in patients who have not responded to standard interferon and ribavirin therapy. Peginterferon alfa-2a is a synthetic, pegylated form of interferon that stimulates the innate antiviral immune response by binding to type 1 interferon receptor, leading to inhibition of viral replication. Ribavirin is a nucleoside analog with broad-spectrum antiviral activity that inhibits viral RNA synthesis and replication. Amantadine is an antiviral agent originally developed for influenza A; its mechanism in hepatitis C treatment remains unclear but may involve interference with viral uncoating or modulation of host immune responses. While this triple therapy has been studied for improving sustained virological response rates in non-responder hepatitis C patients, evidence suggests limited additional benefit from adding amantadine compared to peginterferon alfa-2a plus ribavirin alone[4][6]. The primary indication for this combination has been chronic hepatitis C infection.
