This experimental quadruple combination therapy is designed to achieve a functional cure (HBsAg loss) in patients with chronic hepatitis B. The regimen combines the direct antiviral suppression of tenofovir disoproxil fumarate (a nucleotide analogue) with the immunomodulatory and antiviral effects of peginterferon alfa-2b. To further stimulate the host's immune system against the virus, granulocyte-macrophage colony-stimulating factor (GM-CSF) is utilized as an adjuvant alongside a therapeutic hepatitis B vaccine (HBsAg). This multi-pronged approach aims to break immune tolerance and induce a robust, virus-specific immune response. The therapy has been primarily investigated in pilot studies and investigator-initiated trials at The First Affiliated Hospital, College of Medicine, Zhejiang University.
