Overview

Pegpleranib + anti-VEGF therapy is a **combination treatment** investigated for neovascular (wet) age-related macular degeneration (AMD). Pegpleranib (code name Fovista) is an aptamer that targets platelet-derived growth factor subunit B (PDGF-B), stripping pericytes from abnormal neovascular vessels, thereby making them more susceptible to anti-VEGF therapy, which targets the vascular endothelial growth factor pathway responsible for angiogenesis. The anti-VEGF agent used in combination has commonly been ranibizumab, aflibercept, or bevacizumab. This dual approach aims to maximize vessel regression and visual outcomes compared to anti-VEGF monotherapy, though Phase 3 trials did not meet their prespecified primary efficacy endpoints.
