Pegsebrenatide (NLY01) is a long-acting, PEGylated synthetic peptide that acts as an agonist of the glucagon-like peptide 1 receptor (GLP-1R). It is structurally related to exendin-4 and has been engineered for extended half-life and improved blood-brain barrier penetration. Pegsebrenatide inhibits inflammatory microglial activity and blocks the transformation of astrocytes into neurotoxic A1 astrocytes, thereby reducing dopaminergic cell death. In preclinical models, it ameliorates motor symptoms in Parkinson’s disease (PD) and shows neuroprotective effects in both PD and Alzheimer’s disease models. The drug is currently in clinical development for neurodegenerative diseases[1][2][3][7].
