Pegsitacianine is an intraoperative, intravenous pH-activated fluorescent imaging agent developed for real-time surgical imaging of solid tumors. It is a micellar nanoparticle composed of a hydrophilic polyethylene glycol exterior and a hydrophobic polymethyl methacrylate core covalently linked to the near-infrared (NIR) fluorophore indocyanine green 2. At physiological pH, the fluorophore remains quenched in an "off" state; upon exposure to the acidic tumor microenvironment, the nanoparticle dissociates and releases the fluorophore into an "on" state, enabling visualization with NIR light during surgery. Pegsitacianine is designed as a tumor-agnostic imaging agent compatible with most clinical cameras used for ICG imaging and aims to improve detection of cancerous tissue during cytoreductive surgery across various solid tumors[1][3][5][7].
