Overview

Pegvisomant is a recombinant protein-based drug that acts as a highly selective growth hormone (GH) receptor antagonist. Structurally, it is an analog of human growth hormone that has been modified and conjugated with polyethylene glycol (PEG) polymers to enhance its pharmacokinetic properties. Pegvisomant binds to GH receptors on cell surfaces, blocking the binding and action of endogenous GH, thereby inhibiting downstream signal transduction. This results in decreased production of insulin-like growth factor-I (IGF-I), the primary mediator of GH activity. Pegvisomant is primarily indicated for the treatment of acromegaly in patients who have had an inadequate response to surgery or radiation therapy, or for whom these therapies are not appropriate[1][2][4][5]. It is administered as a subcutaneous injection.
