Overview

Pegylated human recombinant leptin is a modified form of the hormone leptin in which the native human recombinant protein is covalently linked to polyethylene glycol (PEG), increasing its molecular size and serum half-life. Leptin is a key regulator of energy balance, appetite suppression, and body weight control through activation of the leptin receptor (LEPR) in the hypothalamus. Pegylation allows for less frequent dosing by prolonging systemic exposure. In clinical studies, weekly subcutaneous administration of pegylated human recombinant leptin (PEG-OB) was generally well tolerated and led to sustained serum concentrations. It has been investigated primarily for obesity and weight loss in overweight or obese individuals undergoing caloric restriction[2][3][6][7]. The mechanism involves agonism at the leptin receptor, leading to reduced appetite and potential metabolic effects.
