This is a combination therapy consisting of pegylated interferon alpha-2b (a long-acting, polyethylene glycol-conjugated form of recombinant human interferon alpha-2b) and temozolomide (an oral alkylating agent). Pegylated interferon alpha-2b acts as an immunomodulatory cytokine that stimulates the immune system to target tumor cells. Temozolomide is a DNA-methylating agent that induces cytotoxicity in rapidly dividing cancer cells by causing DNA damage and apoptosis. The combination has been investigated primarily for the treatment of advanced or metastatic melanoma and recurrent glioblastoma multiforme, with studies showing moderate antitumor activity and tolerability. The rationale for combining these agents is to leverage both direct cytotoxic effects on tumor cells (temozolomide) and immune-mediated antitumor activity (pegylated interferon alpha-2b)[1][6][9].
