Pegylated liposomal doxorubicin is a formulation of the anthracycline antibiotic doxorubicin encapsulated in long-circulating, polyethylene glycol (PEG)-coated liposomes. The PEG coating (pegylation) protects the drug from detection by the mononuclear phagocyte system and prolongs its circulation time in blood. This unique delivery system allows for enhanced accumulation of the drug in tumor tissues due to leaky tumor vasculature, while reducing exposure to normal tissues. The mechanism of action is through intercalation into DNA and inhibition of topoisomerase II, leading to inhibition of DNA replication and cell death. Pegylated liposomal doxorubicin is primarily used for treating breast cancer, ovarian cancer, multiple myeloma, and HIV-related Kaposi's sarcoma.
