Pegylated liposomal doxorubicin + docetaxel is a combination chemotherapy regimen investigated by the Hebei Medical University Fourth Hospital for the neoadjuvant treatment of breast cancer. This regimen combines pegylated liposomal doxorubicin (PLD), an anthracycline encapsulated in a polyethylene glycol-coated liposome to minimize cardiotoxicity and enhance tumor delivery, with docetaxel, a taxane that promotes microtubule assembly and inhibits depolymerization. Doxorubicin exerts its cytotoxic effect by intercalating into DNA and inhibiting the enzyme topoisomerase II, leading to DNA strand breaks and inhibition of macromolecular synthesis. Docetaxel induces cell cycle arrest at the G2/M phase by stabilizing microtubules. The combination is designed to provide synergistic antitumor activity with a manageable safety profile in patients with locally advanced breast cancer.
