Pegylated recombinant human megakaryocyte growth and development factor (PEG-rHuMGDF) is a truncated, nonglycosylated form of the amino-terminal 163 amino acids of human thrombopoietin (TPO), covalently linked to a 20-kDa polyethylene glycol moiety. It acts as an Mpl-ligand that stimulates the proliferation and differentiation of megakaryocytes in bone marrow, leading to increased platelet production. The drug was developed for the management of severe thrombocytopenia, particularly in patients undergoing chemotherapy or with advanced cancer. Pegylation extends its plasma half-life to approximately 25–35 hours compared to non-pegylated forms. Clinical studies demonstrated dose-dependent increases in platelet counts with low toxicity; platelets produced were morphologically and functionally normal[1][2][3][5][6].
