Drug intelligence / Profile preview
PEI-PIP3 is an experimental polymeric carrier-mediated complex consisting of branched polyethyleneimine (PEI, specifically 25 kDa PEI-25) and the lipid second messenger phosphatidylinositol-3,4,5-trisphosphate (PIP3). Developed by researchers at Ben-Gurion University of the Negev, it is designed as a "second messenger therapeutic" to overcome cellular insulin resistance. PIP3 is a key mediator of insulin signaling, but its high negative charge and membrane impermeability prevent direct cellular uptake. By complexing PIP3 with the cationic polymer PEI, the resulting nanoparticle formulation facilitates trans-membrane delivery. PEI-PIP3 bypasses proximal insulin signaling defects (upstream of endogenous PIP3 generation) to directly activate downstream effectors, stimulating Akt phosphorylation and GLUT4 translocation to restore glucose uptake in insulin-resistant muscle and adipose cells.
