Overview

Pelareorep is a proprietary formulation of the human wild-type serotype 3 Dearing (T3D) strain of reovirus (Respiratory Enteric Orphan virus), developed as an oncolytic virus therapy for cancer. It selectively infects and replicates in cancer cells with activated Ras signaling pathways, leading to direct tumor cell lysis. The replication process also triggers both innate and adaptive immune responses by upregulating interferon expression and pro-inflammatory cytokines, recruiting T cells, natural killer cells, and dendritic cells to the tumor microenvironment. This dual mechanism not only destroys cancer cells directly but also turns immunologically "cold" tumors "hot," making them more susceptible to immune-mediated attack and enhancing the effectiveness of other oncology treatments such as chemotherapy or immune checkpoint inhibitors. Pelareorep is administered intravenously and is being investigated in multiple clinical trials for various cancers including breast cancer (HR+/HER2- and triple-negative), pancreatic cancer, metastatic colorectal cancer, unresectable anal cancer, ovarian cancer, malignant glioma, non-small cell lung cancer (NSCLC), metastatic castration-resistant prostate cancer, multiple myeloma, among others.
