Drug intelligence / Profile preview
pelareorep + atezolizumab + gemcitabine + nab-paclitaxel is an investigational combination regimen being evaluated for the first-line treatment of advanced or metastatic pancreatic ductal adenocarcinoma (PDAC). The regimen combines pelareorep (an intravenously administered, non-genetically modified oncolytic reovirus that selectively replicates in RAS-upregulated tumor cells) with atezolizumab (an anti-PD-L1 immune checkpoint inhibitor) and the standard-of-care chemotherapy doublet gemcitabine and nab-paclitaxel (GnP). Pelareorep acts by directly lysing cancer cells and promoting an inflamed tumor microenvironment, which synergizes with atezolizumab to overcome the highly immunosuppressive nature of pancreatic tumors. This combination has demonstrated promising efficacy and immune conversion activity in Cohort 1 of the Phase 1/2 GOBLET trial, receiving Fast Track Designation from the FDA.
