Drug intelligence / Profile preview
pelareorep + FOLFIRI + bevacizumab is an investigational combination regimen consisting of pelareorep (an intravenously delivered, systemically active, unmodified oncolytic reovirus), FOLFIRI (a chemotherapy regimen of folinic acid, fluorouracil, and irinotecan), and bevacizumab (a recombinant humanized monoclonal antibody targeting vascular endothelial growth factor A, or VEGF-A). Developed by Oncolytics Biotech, this combination is being evaluated as a second-line treatment for patients with RAS-mutant (specifically KRAS-mutant), microsatellite-stable (MSS) metastatic colorectal cancer (mCRC). Pelareorep selectively replicates in cancer cells with activated Ras pathways, leading to direct viral oncolysis and stimulating innate and adaptive anti-tumor immune responses. FOLFIRI provides cytotoxic chemotherapy, while bevacizumab inhibits angiogenesis. Together, the combination aims to enhance anti-tumor efficacy and overcome resistance in immunologically "cold" tumors.
