Pelidotin is a synthetic antimitotic agent used as the cytotoxic payload component in certain antibody-drug conjugates (ADCs) for targeted cancer therapy. Its primary mechanism is microtubule inhibition, disrupting cell division and leading to tumor cell death. Pelidotin is not used as a standalone drug but is conjugated to monoclonal antibodies such as in cofetuzumab pelidotin (PF-06647020, ABBV-647) and micvotabart pelidotin (MICVO), where it acts as the cytotoxic agent delivered directly to tumor cells expressing particular antigens (such as PTK7 or tumor extracellular matrix components). ADCs containing pelidotin have demonstrated preclinical and early clinical antitumor activity across multiple solid tumor types, including ovarian cancer, non-small cell lung cancer, triple-negative breast cancer, and head and neck squamous cell carcinoma[1][2][3][4][5][7][10].
