peluntamig + paclitaxel is an investigational combination therapy under clinical development for the treatment of extensive-stage small cell lung cancer, large cell neuroendocrine carcinoma, extrapulmonary neuroendocrine carcinomas, and neuroendocrine prostate cancer. peluntamig (also known as PT217) is a first-in-class IgG-like bispecific antibody targeting both DLL3 and CD47, intended to enhance immune-mediated attack of neuroendocrine tumors by blocking DLL3 (a Notch pathway component overexpressed on tumor cells) and CD47 (a "don’t eat me" signal protein on cancer cells). paclitaxel is a microtubule stabilizing chemotherapeutic agent (taxane class) that disrupts the normal cycling of microtubule dynamics essential for cell division, leading to apoptosis. This combination is being studied to exploit both direct cytotoxic and immunomodulatory anti-tumor mechanisms[1][3][5][7].
