This combination therapy consists of the PD-1 inhibitor pembrolizumab and the cytotoxic chemotherapies carboplatin and paclitaxel. It is being evaluated by Wake Forest University Health Sciences as a first-line treatment for patients with metastatic head and neck squamous cell carcinoma (HNSCC) who are ineligible for 5-fluorouracil (5-FU). Pembrolizumab is a humanized monoclonal antibody that enhances the immune response against tumor cells by blocking the PD-1/PD-L1 pathway. Carboplatin is a platinum-based agent that induces DNA damage through cross-linking, while paclitaxel is a taxane that inhibits cell division by stabilizing microtubules. This regimen aims to provide an effective alternative to standard-of-care fluorouracil-based therapies for patients with contraindications to 5-FU.
