This chemoimmunotherapy combination regimen is used as a standard-of-care control in clinical trials for squamous non-small cell lung cancer (NSCLC). It consists of the anti-PD-1 monoclonal antibody pembrolizumab, the platinum-based alkylating agent carboplatin, and a taxane chemotherapy agent, which can be either nab-paclitaxel or paclitaxel at the investigator's discretion. Pembrolizumab functions by blocking the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby enhancing T-cell mediated anti-tumor immune responses. Carboplatin induces DNA cross-linking to inhibit replication, while the taxane component (paclitaxel or nab-paclitaxel) stabilizes microtubules to prevent cell division. This specific combination is being utilized by ImmunityBio, Inc. as the reference treatment in the QUILT-2.023 Phase 3 study.
