Overview

This therapeutic regimen is a novel combination therapy being evaluated by Capital Medical University for the treatment of advanced malignant mesothelioma. It consists of the anti-PD-1 monoclonal antibody pembrolizumab (Keytruda), autologous dendritic cell-cytokine induced killer (DC-CIK) cell immunotherapy, and regional hyperthermia. Pembrolizumab functions as a checkpoint inhibitor by blocking the Programmed Cell Death Protein 1 (PD-1) receptor on T-cells, thereby preventing its interaction with ligands PD-L1 and PD-L2 to restore anti-tumor immune activity. The DC-CIK component involves the ex vivo expansion and activation of a patient's own mononuclear cells into a heterogeneous population of immune effectors (including dendritic cells and T-cells) intended to provide direct cytotoxicity and stimulate broader immune responses. Hyperthermia, delivered via the Thermotron RF-8EX device, is used to physically stress tumor cells, potentially increasing their immunogenicity and enhancing the infiltration and efficacy of the immunotherapy components within the tumor microenvironment.
