Overview

This is a **combination regimen** consisting of four agents: - **Pembrolizumab**: a humanized monoclonal antibody (IgG4 kappa) that targets programmed death receptor-1 (**PD-1**) to enhance anti-tumor immune response. - **Doxorubicin**: a small molecule anthracycline chemotherapeutic agent, intercalates DNA and inhibits topoisomerase II, leading to apoptosis of rapidly dividing cells. - **Brentuximab vedotin**: an antibody-drug conjugate composed of an anti-CD30 monoclonal antibody linked to the anti-mitotic agent monomethyl auristatin E, selectively targeting and killing CD30-positive tumor cells. - **Nivolumab**: a human monoclonal antibody that binds PD-1, blocking its interaction with PD-L1/PD-L2, thereby reversing immune suppression in the tumor microenvironment. These agents have individually and in pairs been studied for **relapsed/refractory Hodgkin lymphoma (HL)**, **primary mediastinal large B-cell lymphoma (PMBL)**, **soft tissue sarcoma (STS)**, and **triple-negative breast cancer (TNBC)**, among other cancers. Triple and quadruple regimens containing all four agents are not the current standard of care and are not described in publicly available clinical trials as a single combined regimen.
