pembrolizumab + FOLFIRI

Pembrolizumab + FOLFIRI is a combination regimen composed of **pembrolizumab**, an immune checkpoint inhibitor monoclonal antibody targeting PD-1, and **FOLFIRI**, a chemotherapeutic regimen consisting of folinic acid (leucovorin), fluorouracil (5-FU), and irinotecan. Pembrolizumab potentiates antitumor immunity by blocking the PD-1 receptor, thereby enhancing T-cell–mediated responses against tumor cells. FOLFIRI includes agents that disrupt DNA synthesis and replication in rapidly dividing cancer cells, primarily used in metastatic colorectal cancer and under investigation for other gastrointestinal malignancies. The rationale for combining pembrolizumab with FOLFIRI is that chemotherapy may augment immune responses, potentially providing greater tumor control than either therapy alone, especially in patients with advanced or previously treated tumors such as metastatic microsatellite stable/mismatch repair proficient colorectal cancer[1][2][4][9].
