Drug intelligence / Profile preview
Pembrolizumab + irinotecan is an investigational combination regimen consisting of the anti-PD-1 monoclonal antibody pembrolizumab (Keytruda) and the topoisomerase I inhibitor chemotherapy agent irinotecan. This combination is being evaluated in clinical trials, including a Phase 1b/2 trial sponsored by Yonsei University for the treatment of metastatic or recurrent malignant melanoma, and a Phase 2 trial for high-grade neuroendocrine cancer. The therapeutic rationale is to combine the cytotoxic, DNA-damaging effects of irinotecan with the immune checkpoint blockade of pembrolizumab to enhance anti-tumor efficacy and potentially overcome resistance in advanced solid tumors.
