Overview

Pembrolizumab + lenvatinib is a fixed combination regimen of two anticancer agents used for the treatment of certain advanced cancers. Pembrolizumab is a humanized monoclonal antibody that targets the programmed cell death protein 1 (PD‑1) receptor, acting as an immune checkpoint inhibitor to enhance T-cell–mediated antitumor responses. Lenvatinib is an orally available small molecule tyrosine kinase inhibitor that targets multiple receptors involved in tumor angiogenesis and proliferation, including vascular endothelial growth factor receptors (VEGFR1–3), fibroblast growth factor receptors (FGFR1–4), platelet-derived growth factor receptor alpha (PDGFRα), RET, and KIT. The combination has demonstrated synergistic antitumor activity by modulating both the immune microenvironment and tumor vasculature. It is approved for first-line treatment of advanced renal cell carcinoma and for advanced endometrial carcinoma not MSI-H or dMMR after prior systemic therapy in patients who are not candidates for curative surgery or radiation[2][3][4][5][6][9].
