Overview

This is a combination regimen consisting of **pembrolizumab** (a monoclonal antibody targeting programmed death-1 [PD-1]), **MK-4830** (a monoclonal antibody targeting immunoglobulin-like transcript 4 [ILT4]), and **lenvatinib** (a small molecule multikinase inhibitor targeting VEGFR1-3, FGFR1-4, PDGFRα, RET, and KIT). The combination is being evaluated in clinical trials for advanced solid tumors and renal cell carcinoma. Pembrolizumab blocks PD-1 on T cells, releasing immune suppression and promoting anti-tumor immune activity. MK-4830, an investigational anti-ILT4 antibody, aims to further enhance the immune response by inhibiting myeloid cell-mediated suppression of anti-tumor immunity. Lenvatinib inhibits multiple tyrosine kinases involved in tumor growth, angiogenesis, and progression. This triple combination seeks to synergistically enhance anti-tumor activity via immune checkpoint inhibition, myeloid cell modulation, and targeted blockade of growth factor signaling pathways[1][2][3][4].
