Overview

This triplet combination therapy consists of the PD-1 inhibitor pembrolizumab, the pan-HER tyrosine kinase inhibitor neratinib, and the microtubule-stabilizing cytotoxic agent paclitaxel. It is being investigated by Yonsei University as a second-line treatment for patients with recurrent or advanced gastric cancer who harbor somatic mutations in the HER family (EGFR, HER2, HER3, HER4) or HER2 amplification/overexpression. The regimen aims to leverage the synergistic effects of immune checkpoint blockade, targeted inhibition of the HER signaling pathway, and traditional chemotherapy to overcome resistance and improve clinical outcomes in this molecularly defined subpopulation.
