Overview

This is a **combination immunotherapy** regimen comprising three monoclonal antibodies: **pembrolizumab**, a programmed cell death protein 1 (PD-1) inhibitor; **quavonlimab**, a cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) inhibitor; and **vibostolimab**, a T cell immunoreceptor with Ig and ITIM domains (TIGIT) inhibitor. Each antibody blocks a distinct immune checkpoint pathway, collectively aiming to restore and enhance T cell-mediated anti-tumor immunity by simultaneously preventing immune exhaustion (PD-1), suppressing regulatory T cell activity (CTLA-4), and antagonizing innate and adaptive immune suppression (TIGIT). Development is led by **Merck**. The triplet is principally tested in advanced solid tumors, including melanoma, non-small cell lung cancer (NSCLC), and others, within the KEYMAKER and KEYVIBE clinical trial programs[2][4].
