Overview

This is a combination therapy under investigation for the treatment of estrogen receptor-positive (ER+) breast cancer. The regimen combines pembrolizumab (an immune checkpoint inhibitor), tamoxifen (a selective estrogen receptor modulator), and vorinostat (a histone deacetylase inhibitor). The rationale is to overcome hormone therapy resistance by epigenetic priming with vorinostat, which may enhance the immune response to pembrolizumab while maintaining endocrine control with tamoxifen. Clinical trials are evaluating the safety and efficacy of this combination in patients with advanced or metastatic ER+ breast cancer who have progressed on prior therapies[1][4][7].
