Pembrolizumab + tivumecirnon is a combination therapy consisting of pembrolizumab, a PD-1 inhibitor, and tivumecirnon (FLX475), a selective CCR4 antagonist. Tivumecirnon is designed to block the migration of regulatory T cells (Tregs) into tumors, thereby preventing immunosuppression in the tumor microenvironment. When combined with pembrolizumab, which blocks the PD-1 pathway, this dual approach aims to enhance anti-tumor immunity by both preventing Treg-mediated immunosuppression and reinvigorating exhausted T cells. The combination has shown promising activity in several cancer types, particularly in virus-associated cancers such as EBV-positive gastric cancer and HPV-positive head and neck squamous cell carcinoma.
