Pembrolizumab + tretinoin is a **combination therapy** consisting of **pembrolizumab**, an immune checkpoint inhibitor (anti-PD-1 monoclonal antibody), and **tretinoin** (all-trans retinoic acid, ATRA), a vitamin A derivative. Pembrolizumab blocks the PD-1 pathway, thereby promoting T-cell-mediated anti-tumor immunity. Tretinoin targets myeloid-derived suppressor cells (MDSCs), reducing their frequency and suppressive function, thus enhancing anti-tumor immune responses when combined with pembrolizumab. This combination was investigated primarily for advanced melanoma. Clinical studies showed high response rates and durable complete responses in anti-PD-1 naïve advanced melanoma patients, with a favorable safety profile[1][2][3][5].
