Overview

Pembrolizumab + vicriviroc is a combination therapy evaluated for the treatment of advanced or metastatic microsatellite stable (MSS) or mismatch repair proficient (pMMR) colorectal cancer. Pembrolizumab is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor to enhance anti-tumor immune responses. Vicriviroc is a small-molecule antagonist of the C-C motif chemokine receptor 5 (CCR5), which blocks signaling involved in tumor progression and immune cell trafficking. The combination was studied in phase 2 clinical trials but demonstrated limited antitumor activity in this patient population; toxicity was manageable[1][2][3][7].
