Overview

Pembrolizumab + vorinostat is a combination therapy under investigation for the treatment of various cancers, particularly relapsed or refractory classical Hodgkin lymphoma (cHL), primary mediastinal B-cell lymphoma (PMBCL), diffuse large B-cell lymphoma, follicular lymphoma, and metastatic non-small cell lung cancer (NSCLC)[1][3][4][5][6]. Pembrolizumab is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor on T cells, blocking its interaction with PD-L1 and PD-L2 to enhance anti-tumor immune responses. Vorinostat is a small molecule histone deacetylase inhibitor (HDACi) that increases tumor immunogenicity by upregulating genes involved in antigen presentation and T-cell recruitment[2][4]. The combination aims to improve response rates by both enhancing immune activation via PD-1 blockade and increasing tumor visibility to the immune system through epigenetic modulation. Clinical trials have shown promising activity in patients with cHL refractory to prior PD-1 blockade as well as notable efficacy in PMBCL[1][3][7].
