Pembrolizumab-activated DC-CIK cells represent an adoptive cell therapy combining dendritic cell-stimulated cytokine-induced killer cells (DC-CIK) that are pretreated in vitro with pembrolizumab, a monoclonal antibody targeting PD-1, before infusion to the patient. This approach aims to block PD-1-mediated inhibitory signaling in the DC-CIK cells, thereby enhancing their cytotoxic immune response and proliferation against tumor cells[1][3][4]. Mechanistically, the PD-1 blockade via pembrolizumab prevents immune exhaustion, increases immune cell infiltration at the tumor site, and promotes the production of perforin, granzyme B, and IFN-γ—key molecules for tumor cell lysis[1]. The combination has shown improved antitumor efficacy in preclinical models of hepatocellular carcinoma (HCC) as well as early-phase studies in lung and other advanced cancers[1][3][4].
