pemetrexed + carboplatin + placebo is a combination chemotherapy regimen used as a control arm in clinical research for advanced non-small cell lung cancer (NSCLC). The regimen consists of pemetrexed, a multi-targeted antifolate that disrupts cell replication by inhibiting enzymes such as thymidylate synthase (TS), dihydrofolate reductase (DHFR), and glycinamide ribonucleotide formyltransferase (GARFT), and carboplatin, a platinum-based alkylating agent that forms DNA cross-links to inhibit DNA synthesis and induce apoptosis. In the context of the University Hospital Muenster trial (NCT00441532), this doublet was administered with a placebo to evaluate the incremental benefit of adding the multi-kinase inhibitor sorafenib to standard first-line therapy for patients with stage IIIb or IV NSCLC.
