Overview
This regimen is a combination therapy used primarily for the treatment of non-squamous non-small cell lung cancer (NSCLC) and malignant pleural mesothelioma. It consists of the antifolate chemotherapeutic agent pemetrexed, combined with either carboplatin or cisplatin, both platinum-based chemotherapies. Sintilimab, a monoclonal antibody targeting PD-1, may be added to enhance immune-mediated anti-tumor activity. Pemetrexed inhibits multiple folate-dependent enzymes involved in nucleotide synthesis, thereby blocking DNA replication in rapidly dividing cells. Carboplatin and cisplatin form DNA crosslinks that prevent cell division and induce apoptosis. Sintilimab blocks the PD-1 receptor on T-cells to restore immune response against tumor cells[2][4][6][7][8].
