This is a combination regimen of three anticancer drugs—pemetrexed, gemcitabine, and bevacizumab—used primarily in the treatment of advanced non-small cell lung cancer (NSCLC). Pemetrexed is an antifolate chemotherapy agent that inhibits several folate-dependent enzymes involved in nucleotide synthesis, thereby disrupting DNA replication and cell division[8]. Gemcitabine is a nucleoside analog that interferes with DNA synthesis by incorporating into DNA strands and inhibiting ribonucleotide reductase. Bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis required for tumor growth. The combination has been investigated as first-line therapy for advanced NSCLC, showing activity and tolerability in phase II clinical trials[1][2]. Each drug contributes distinct mechanisms to inhibit tumor proliferation and survival.
