Pemetrexed + ramucirumab is a combination therapy consisting of pemetrexed, an antifolate chemotherapy agent, and ramucirumab, a fully human monoclonal antibody that specifically targets vascular endothelial growth factor receptor-2 (VEGFR-2). This combination has been investigated for the treatment of advanced/metastatic non-small cell lung cancer (NSCLC). Ramucirumab works by binding with high affinity to the extracellular domain of VEGFR-2, inhibiting the binding of VEGF receptor ligands VEGF-A, VEGF-C, and VEGF-D. This prevents VEGF-stimulated receptor phosphorylation and downstream ligand-induced proliferation, permeability, and migration of human endothelial cells, ultimately inhibiting angiogenesis.
